    Product description 

    Rectal suppositories Pentasa are indicated for ulcerative proctitis.

    Compound

    Active ingredient: mesalazine;
    1 suppository contains mesalazine 1000 mg;
    Excipients: povidone, macrogol 6000, magnesium stearate, talc.

    Contraindications

    Hypersensitivity to salicylates, the active substance or any of the excipients. Severe violations of the liver and / or kidneys. Peptic ulcer of the stomach or duodenum. Hemorrhagic diathesis.

    Mode of application

    Adults - 1 g 1-2 times a day.
    Do not reduce doses in elderly patients.
    Immediately before the introduction of the suppository, it is recommended to empty the intestines.
    To ensure hygienic manipulation, a rubber fingertip should be used.
    The suppository is inserted as deep as possible into the rectum. The suppository should be kept in the rectum for as long as possible to achieve the maximum therapeutic effect.
    If the lesion is widespread or in cases where the response to oral treatment is delayed, suppositories can be used simultaneously with tablets.
    The duration of use is determined by the doctor.

    Overdose

    There is only limited clinical experience with an overdose of Pentasa, which does not indicate the presence of renal or hepatic toxicity. There is no specific antidote, treatment should be symptomatic and supportive. There have been reports of patients using daily doses of the preparation up to 8 g for a month without any side effects.
    Given the dosage form of mesalazine, the risk of overdose is low.
    In general, the symptoms should correspond to those of salicylic acid poisoning: acid-base intoxication, hyperventilation of the lungs, dehydration caused by sweating and vomiting, hypoglycemia.
    Treatment of overdose: with acidosis or alkalosis - restoration of acid-base and electrolyte balance; with dehydration - rehydration; with hypoglycemia - the use of glucose. Additionally, carry out intravenous administration of electrolytes to increase diuresis. Careful monitoring of kidney function.

    Side effects

    The most common adverse reactions observed during clinical studies were diarrhea, nausea, abdominal pain, headache, vomiting and rash.
    Hypersensitivity reactions and preparation fever are sometimes observed.
    After rectal administration, local reactions such as itching, rectal discomfort and urge to defecate are possible.

    Interaction

    Simultaneous treatment with Pentasa and azathioprine, 6-mercaptopurine or thioguanine in studies led to an increase in the frequency of myelosuppressive effects; probably, there is an interaction between these preparations. The mechanism of this interaction has not been fully elucidated. It is recommended to regularly monitor the level of leukocytes and adjust the dose of thiopurine accordingly.
